Genetic diversity within Mycobacterium tuberculosis complex impacts on the accuracy of genotypic pyrazinamide drug-susceptibility assay

چکیده
We agree with Pholwat et al. [1] that fast genotypic methods will play an increasingly prominent role in drug-susceptibility testing (DST) for the Mycobacterium tuberculosis complex (MTBC) [2]. They reported on the evaluation of a high-resolution melt assay (HMR) to detect pyrazinamide (PZA) resistance, which has the advantage of speed and simplicity. Two of 96 test isolates were PZA susceptible but contained a synonymous pncA

Comparison of phenotypic and genotypic methods for pyrazinamide susceptibility testing with Mycobacterium tuberculosis.
Mycobacterium tuberculosis converts pyrazinamide to its active form by using the enzyme pyrazinamidase. This enzyme is coded for on the pncA gene, and mutations in the pncA gene result in absence of active enzyme, conferring resistance to the drug pyrazinamide. We investigated 27 strains of Mycobacterium tuberculosis suspected of being multidrug resistant. Each isolate was tested for susceptibi...
متن کاملGenotypic determination of Mycobacterium tuberculosis antibiotic resistance using a novel mutation detection method, the branch migration inhibition M. tuberculosis antibiotic resistance test.
A novel method for the detection of any alteration within a defined sequence has recently been demonstrated (A. Lishanski, N. Kurn, and E. F. Ullman, Nucleic Acids Res. 28:E42, 2000; A. Lishanski, Clin. Chem. 46:9, 2000). Essential to this method are the generation of partial duplexes that are capable of forming four-stranded structures and the ability to detect inhibition of branch migration i...
متن کاملpncA GENE SEQUENCE ANALYSIS FOR PYRAZINAMIDE RESISTANCE IN MYCOBACTERIUM TUBERCULOSIS FROM HIGH-INCIDENCE SETTING
Standard culture-based drug susceptibility testing (DST) of Mycobacterium tuberculosis (MTB) to pyrazinamide (PZA) is difficult to perform. PZA is a prodrug that has to be converted to its active form pyrazinoic acid (POA) by pyrazinamidase (PZase) enzyme, encoded by pncA gene and loss of PZase activity is associated with PZA resistance. To further define genetic basis of PZA resistance and det...
متن کاملComparison of phenotypic and genotypic methods for pyrazinamide susceptibility testing.
OBJECTIVES To evaluate Pyrazinamide (PZA) susceptibility results obtained by phenotypic MGIT 960 TB system against enzymatic Pyrazinamidase assay and genotypic pncA gene sequencing. To find the prevalence of infections caused by M. bovis in PZA resistant M. tuberculosis complex isolates. METHODS 33 consecutive PZA resistant and 30 consecutive PZA susceptible isolates reported for PZA suscepti...
متن کاملRapid detection of Mycobacterium tuberculosis and pyrazinamide susceptibility related to pncA mutations in sputum specimens through an integrated gene-to-protein function approach.
Testing the pyrazinamide (PZA) susceptibility of Mycobacterium tuberculosis isolates is challenging. In a previous paper, we described the development of a rapid colorimetric test for the PZA susceptibility of M. tuberculosis by a PCR-based in vitro-synthesized-pyrazinamidase (PZase) assay. Here, we present an integrated approach to detect M. tuberculosis and PZA susceptibility directly from sp...
